Clinical Significance
Lipase is an enzyme that hydrolyses glycerol esters and long-chain fatty acids. Enzyme and its cofactor colipase are produced in the pancreas, lipase is also secreted in small amounts by the salivary glands, pulmonary and intestinal mucosa. Bile acids and colipase are hidden in them to form complexes with lipids and lipase will bind on the substrate/water. Lipase determination is used for the investigation of disorders of the pancreas. In acute pancreatitis, lipase concentrations exceed the maximum reference limit by 20-50 times within 4-8 hours after the onset of abdominal pain with a peak in the 24 hours and a decrease in the following 8 to 14 days. Elevated lipase values can also be observed in chronic pancreatitis and obstructions of the duct of the pancreas.
Test Principle
The colorimetric substrate, 1,2-O-Dilauryl-rac-glycero-3-glutaric-(6'methyl-resorufin)ester acid, is cleaved by pancreatic lipase and the resulting dicarboxylic acid ester is hydrolysed under the alkaline test conditions, generating the chromogen methylresorufin. The kinetics of colour development are monitored at 580 nm and are proportional to the activity of lipase in the sample.
Stability & Storage
Store at 2 - 8°C away from direct light. Stored as described above, the reagents are stable until the expiry date stated on the label. A slight variation in the composition of the reagents can occur from batch to batch without affecting the test results. Reagents are liquid and ready to use. The stability of the reagents after opening is 60 days if kept closed at 2 - 8°C protected from direct light. The solutions must be clear without noticeable precipitates. In use, avoid bacterial contamination. Attention: the R2 reagent is a microemulsion. Apparent precipitation may occur, with the formation of a red circle at the base of the bottle. This behaviour should not be alarming and delicate reversing mixing is sufficient before using the reagent.
